The enrichment is gentle and fast and can be performed in less than three hours. By tying up water molecules, the reagent forces less-soluble components such as vesicles out of solution, allowing them to be collected by a short, low-speed centrifugation. The isolated intact virus can be used for functional studies, in contrast to current SARS-CoV-2 purification methods based on extracting RNA for detection purposes. Use twice as much virus sample as precipitation reagent (e.g., use 2 mL virus sample per 1 mL reagent).
1. Add precipitation reagent to the virus sample and incubate for 2 hours at 2–8°C.
2. Centrifuge for 30 minutes at 10,000 x g.
3. Remove supernatant and keep the pellet with the enriched virus.
The enriched virus is now ready for any downstream analysis and can be used directly in a PCR reaction without any further nucleic acid extraction.
