123Count eBeadsは、フローサイトメトリーによる細胞や他の粒子の絶対数のカウント用に意図されています。励起源に適合する青色(488 nm)および紫色(405 nm)の色素がカプセル化され、約500 nm~750 nmの蛍光を発する7 μmの微粒子です。これらの微粒子は、2 mMのアジ化ナトリウムを添加した、既知濃度0.05%のTween-20で提供されます。フローサイトメーターで細胞とビーズが浮遊する混合物の測定値を慎重に収集することで、細胞のレシオメトリック測定を可能にします。

I am using counting beads to count cells, but I cannot find the beads on my scatter plots. What do I do?

The first thing to do is check your threshold and see if it is set on forward scatter. If so, the beads are probably being excluded by the threshold. Reducing the threshold setting should reveal your beads.

I want to count my cells using flow cytometry. How do I do this?

Cell counting using flow cytometry can be accomplished by adding an internal microsphere counting standard to the flow cytometric sample. The number of reference beads that are collected reflects a known volume. This allows you to calculate cell concentration.

What does yield and purity mean in reference to the MagniSort kits? How are each of these parameters calculated?

Yield is the percentage of target cells harvested based on the number of cells theoretically available in the starting population. To calculate yield, stain a sample of unsorted and sorted cells with antibodies to identify the target cells, and count the total number of target cells using 123count eBeads (Cat. No. 01-1234-42) on the cytometer. Yield = # of sorted target cells, # of target cells unsorted.

Purity is the percentage of cells harvested that are target cells. Purity can be directly generated by flow cytometric analysis of sorted samples. To obtain purity, stain the sorted cells with antibodies to identify the target cells and analyze by flow cytometry. Gate on live, single cells, and subsequently gate on the target cell population. Purity is the percentage of target cells in the live, single-cell gate.
