Pierce Glutathione Coated Plates are coated with intact glutathione (GSH) and pre-blocked for use in capture and detection assays involving glutathione-S-transferase (GST) fusion proteins. These coated plates can be used in the analysis of cell lysates to determine the presence and concentration of the GST-fusion protein produced in an expression system. Plates can also be used to capture and coat fusion proteins via their GST tag for subsequent binding assays involving the recombinant protein. As the plates are pre-blocked, initial purification of the cell lysates is not necessary.

My initial primary culture is contaminated, what can I do?

Contamination of primary tissue could possibly be carried over to culture preparation. Change the media and treat with antibiotics to eliminate the contaminants. To prevent this in the future, wash tissue pieces several times in a balanced salt solution containing a higher concentration of antibiotics and antimycotics before starting culture.

What is the detection limit of proteins with Pierce Glutathione Coated Plates?

The background is extremely low when using the Pierce Glutathione Coated Plates. This results in a high signal-to-noise ratio and a low detection limit which in general means that the detection limit (OD 0.5) is 3 ng per well (100 µL).

How long are Pierce Glutathione Coated Plates stable?

The Pierce Glutathione Coated Plates are extremely stable over long periods of time, if stored at room temperature (20-25 degrees C). We guarantee 12 mths of shelf life in unopened foil package after the date of shipment.

Can I determine the concentration of an unknown GST-tagged protein?

To determine the approximate concentration of a GST-tagged protein, a standard curve should be generated using purified GST added in different concentrations to the wells of the plates. Please note that it is only the approximately concentration that can be determined since not 100% of the GST-fusion proteins will be bound due to denaturing or steric hindrance to GSH.

Can I whirl-mix solutions containing GST proteins?

No. Do not whirl-mix a solution containing GST-tagged proteins. The CV will increase due to slight degradation of the GST protein.
