Applied Biosystems TaqMan Gene Expression assays are used for quantitative real-time PCR or digital PCR analysis of gene expression and consist of a pair of unlabeled PCR primers and a TaqMan probe with a dye label (FAM, VIC, ABY, JUN, and Cyanine 5) on the 5' end and a minor groove binder (MGB) and non-fluorescent quencher (NFQ) on the 3' end.

TaqMan® probesの推奨される保存温度は何℃ですか?

蛍光色素がラベルされているプローブは直接光を当てることを避けてください。そのためプローブが入っているチューブはアルミホイルで包んだ状態で箱に入れて-20℃での保存をお奨めします。

凍結融解を繰り返すことを避けるために小分け分注して保存することをお奨めします。実験作業の時間程度であれば遮光の必要はありません。

ヒトのbeta actinを検出するためのキットでリポーター蛍光色素としてFAM以外のものがラベルされているものは販売していますか?

TaqMan Gene Expression Assayで解析したPCR産物をアガロース電気泳動でチェックすることは可能ですか?

はい、PCR産物をアガロースゲル電気泳動でチェックすることが可能です。ただしTaqMan Universal PCR Master Mix (AmpErase UNG含有, Cat. No. 4304437)のようにAmpErase UNGが含有しているMaster Mixを使用した場合は、PCR反応後、直ちに電気泳動を行ってください。反応液中に微量に残存するAmpErase UNGがdUTPを取り込んだPCR産物を分解してしまうためです。なおTaqMan Gene Expression Assayで増幅されたPCR産物のアンプリコンサイズは、大体50~150bpです。各アッセイのアンプリコンサイズは、website上のアッセイの詳細情報のページに記載があります。アッセイ検索ページからAssay ID番号で検索を行い、検索結果の画面上の"Assay Deteails"ボタンをクリックして頂ければ、詳細情報が確認できます。
