The ExpiCHO Expression System Kit combines the power of rapid, ultra–high-yield transient protein production in suspension culture with the benefit of expression in Chinese hamster ovary (CHO) cells. It is based on high-density culture of ExpiCHO-S cells in ExpiCHO Expression Medium. Transient expression is powered by ExpiFectamine CHO transfection reagent, designed specifically for high-density CHO suspension culture, in combination with a specialized transfection enhancer and culture feed.

All components work together synergistically with expression yields of 1–3 g per L of transfected culture demonstrated for some antibody and non-antibody proteins.

The ExpiCHO Expression System enables:

  • Milligram to multigram yields of recombinant protein per liter of culture in 5–14 days post-transfection
  • Robust and reproducible transfection results, for greater confidence in results and ability to use CHO cells from research to discovery
  • Transfection efficiency of > 90% for high-density suspension CHO cell culture, with matching transfection enhancer and culture feed to boost transfection performance and protein expression
  • Use of one-third to one-half less plasmid DNA than other transfection reagents
  • Flexible, reliable culture and transfection protocols with multiple options that readily fit into current transient expression workflows
  • Scalable expression for culture formats from <1 mL in deep well plates up to 5 L when using Nalgene angled-bottom shake flasks

The components included in the ExpiCHO Expression System Kit are: 2 vials of frozen ExpiCHO-S cells, 1 L of ExpiCHO Expression Medium, 1 ExpiFectamine CHO Transfection Kit sufficient to transfect 1 L of culture, OptiPRO SFM, and an antibody-expressing positive control vector.

ExpiCHO-S cells, ExpiCHO Expression Medium, ExpiFectamine CHO Transfection Kit, and OptiPRO SFM may also be purchased separately.

All components of the system are animal origin–free.

When subculturing ExpiCHO the day before a transfection to 3-4 x 10E6/mL, would you recommend pelleting the cells and resuspending them in completely fresh media, or simply replacing some of the media to split them down to the desired cell density?

Before transfecting ExpiCHO cells, there is no need to change out the media during the subculturing. This would become expensive and the conditioned media is not necessary.

For how many passages would you recommend using ExpiCHO cells before replacing them with new cells that were frozen at earlier passages?

For optimal performance with respect to protein production, we recommend thawing ExpiCHO cells and letting them grow for 2-3 passages prior to transfection. Overall, we do not recommend using the cells beyond ˜20 passages.

With the ExpiCHO Expression System, can I shake faster or slower than recommended?

We have provided optimal shake speeds for various formats. For most flasks, there is a drop-off in expression when you go too fast or slow because of cell shear stress or insufficient aeration. The effect is sharper in plates where there is a sharper transition between static and moving fluid in wells.

How many days should I wait before harvesting my protein with the ExpiCHO system?

Optimal time to harvest protein will depend on the specific properties of the protein being expressed and the protocol chosen. Typical harvest times to reach maximum titers for the various protocols are as follows:
- Standard Protocol: 8-10 days post-transfection
- High Titer Protocol: 10-12 days post-transfection
- Max Titer Protocol: 12-14 days post-transfection
