5-CFDA, AM is a cell-permeant esterase substrate that can serve as a viability probe that measures both enzymatic activity, which is require to activate its fluorescence, and cell-membrane integrity, which is required for intracellular retention of their fluorescent product. The electronically neutral AM ester of CFDA can be loaded into cells at lower concentrations than CFDA. Upon hydrolysis by intracellular esterases, this AM ester also yields carboxyfluorescein. As compared with fluorescein, carboxyfluorescein contains extra negative charges and is therefore better retained in cells.

Abstract
Calcein: a novel marker for lymphocytes which enter lymph nodes.
Authors:Weston SA, Parish CR
Journal:Cytometry
PubMed ID:1451604
Previous studies have identified unique cell surface antigens which are associated with the specific binding of lymphocytes to high endothelial venules (HEV). Evidence is presented in this paper which demonstrates that uptake of the fluorescent dye calcein by lymphocytes represents an additional marker for the lymph node homing subpopulation of ... More
Mechanistic studies of lantibiotic-induced permeabilization of phospholipid vesicles.

'Nisin is a cationic polycyclic bacteriocin secreted by some lactic acid bacteria. Nisin has previously been shown to permeabilize liposomes. The interaction of nisin was analyzed with liposomes prepared of the zwitterionic phosphatidylcholine (PC) and the anionic phosphatidylglycerol (PG). Nisin induces the release of 6-carboxyfluorescein and other small anionic fluorescent ... More
Ability of 16 priority PAHs to be photocytotoxic to a cell line from the rainbow trout gill.
'Sixteen polycyclic aromatic hydrocarbons (PAHs) were screened for their ability to be photocytotoxic to a cell line from the rainbow trout gill, RTgill-W1. PAHs could be divided into one of three groups: incapable of being photocytotoxic, able to be both photocytotoxic and directly cytotoxic, or capable of being only photocytotoxic. ... More
Cell death-associated translocation of plasma membrane components induced by CTL.

'In the very early stages of target cell apoptosis induced by CTL, we found that fluorescence of labeling probes of the target plasma membrane, such as N-(3-triethylammoniumpropyl)-4-(p-dibutylaminostyryl)pyridin ium dibromide (FM1-43), was translocated into intracellular membrane structures including nuclear envelope and mitochondria. This translocation was associated with the execution of CTL-mediated ... More
Comparison of four different colorimetric and fluorometric cytotoxicity assays in a zebrafish liver cell line.
Authors:Bopp SK, Lettieri T,
Journal:BMC Pharmacol
PubMed ID:18513395
'BACKGROUND: A broad spectrum of cytotoxicity assays is currently used in the fields of (eco)toxicology and pharmacology. To choose an appropriate assay, different parameters like test compounds, detection mechanism, specificity, and sensitivity have to be considered. Furthermore, tissue or cell line can influence test performance. For zebrafish (Danio rerio), as ... More
