よくあるご質問(FAQ)

Can I use Hoechst dyes to do both DNA quantitation and cell number counting in plates?

Yes, this can be done in a 96-well plate-based format. Hoechst dyes will label all cells, live or dead (or even fixed). This requires the use of a standard curve of known cell numbers (up to 100,000 cells per well) and a standard curve using purified DNA, , up to 10 µg/mL. We offer a kit for this: FluoReporter Blue Fluorometric dsDNA Quantitation Kit, Cat. No. F2962.

Abstract
Dominant-negative C/EBP disrupts mitotic clonal expansion and differentiation of 3T3-L1 preadipocytes.
Authors:Zhang JW, Tang QQ, Vinson C, Lane MD
Journal:Proc Natl Acad Sci U S A
PubMed ID:14688407
'Hormonal induction of growth-arrested 3T3-L1 preadipocytes rapidly activates expression of CCAAT/enhancer-binding protein (C/EBP) beta. Acquisition of DNA-binding activity by C/EBPbeta, however, is delayed until the cells synchronously enter the S phase of mitotic clonal expansion (MCE). After MCE, C/EBPbeta activates expression of C/EBPalpha and peroxisome proliferator-activated receptor gamma, which then ... More
In vivo imaging platform for tracking immunotherapeutic cells.

Journal:Nat Biotechnol
PubMed ID:16041364
Cellular therapeutics show great promise for the treatment of disease, but few noninvasive techniques exist for monitoring the cells after administration. Here we present a magnetic resonance imaging (MRI) technology that uses perfluoropolyether (PFPE) agents to track cells in vivo. Fluorine MRI selectively images only the labeled cells, and a ... More
ATP regulates the differentiation of mammalian skeletal muscle by activation of a P2X5 receptor on satellite cells.
Authors:Ryten M, Dunn PM, Neary JT, Burnstock G
Journal:J Cell Biol
PubMed ID:12135987
ATP is well known for its role as an intracellular energy source. However, there is increasing awareness of its role as an extracellular messenger molecule (Burnstock, 1997). Although evidence for the presence of receptors for extracellular ATP on skeletal myoblasts was first published in 1983 (Kolb and Wakelam), their physiological ... More
5-HT2A receptor induces ERK phosphorylation and proliferation through ADAM-17 tumor necrosis factor-alpha-converting enzyme (TACE) activation and heparin-bound epidermal growth factor-like growth factor (HB-EGF) shedding in mesangial cells.
Authors:Göoz M, Göoz P, Luttrell LM, Raymond JR
Journal:J Biol Chem
PubMed ID:16737974
In this study, we present multiple lines of evidence to support a critical role for heparin-bound EGF (epidermal growth factor)-like growth factor (HB-EGF) and tumor necrosis factor-alpha-converting enzyme (TACE) (ADAM17) in the transactivation of EGF receptor (EGFR), ERK phosphorylation, and cellular proliferation induced by the 5-HT(2A) receptor in renal mesangial ... More
