Why am I getting negative fluorescence values with my Qubit Assays?
Negative fluorescence is a physical impossibility. It is an artifact from software autocorrecting for background signal. This means your reader is picking up and subtracting out background light at the cost of your data. Make sure to do a buffer-only control and assess the type of signal. You may need to switch to a different plate.
I have a Quant-iT RNA Assay Kit and want to use it with the Qubit Fluorometer. Can I?
Yes, the manual has directions for this application.
Will these Quant-iT RNA kits work for siRNA or microRNA?
No, they are not accurate for such small fragments. The Quant-iT microRNA Assay Kit and Qubit microRNA Assay Kit are good tools for this application.
Can the Quant-iT RNA assay be used to quantitate RNA labeled with digoxigenin?
Yes, the tag does not interfere with the signal.
What are the linear ranges of the Quant-iT RNA kits?
The linear ranges are:
