Gibco FreeStyle F17 Expression Medium is a chemically defined, protein-free, serum-free medium developed to support the growth and transfection of HEK293 cells in suspension. The addition of 4–8 mM L-glutamine or GlutaMAX supplement is required before use.

Little or no required adaptation
293-F, 293-H, and FreeStyle 293-F cells in suspension can be cultured directly into FreeStyle F17 Expression Medium without adaptation. Cells adapted to other commercially available serum-free media can be subcultured directly into FreeStyle F17 medium, usually without any further adaptation. Cells usually require adaptation from serum-containing formulations.

Growth and transfection of HEK293 cells
FreeStyle F17 Expression Medium supports 293-F cells at a peak density of 6–7 x 106 viable cells per mL. Cells cultured in FreeStyle F17 medium can be transfected using a wide variety of transfection reagents, including lipid-based transfection reagents, such as 293fectin and Freestyle MAX Reagent.

Scalability in spinner flasks and bioreactors
Production using FreeStyle F17 Expression Medium can be scaled up in spinner flasks or bioreactors. The appropriate spinner or impeller speed and seeding density should be optimized for each system. Depending on the impeller design and speed, it may be necessary to supplement FreeStyle F17 medium with additional Pluronic F-68 to avoid sheer stress in the culture.

We typically keep the culture going for 7-10 days posttransfection. After that, protein expression may drop.

What is the difference between FreeStyle 293 Expression Medium and FreeStyle F17 Expression Medium?

FreeStyle F17 Expression Medium was designed from a different base formulation than FreeStyle 293 Expression Medium. FreeStyle F17 medium shows equal or better performance than the FreeStye 293 medium for certain cell lines and proteins. It is capable of supporting the growth of 293 and CHO cells and offers a wider variety and selection of transient cell culture media options.
