The Gibco AAV-MAX Helper-Free AAV Production System is a scalable, high-titer platform for suspension AAV production. It is based on a high-density suspension culture of clonal 293F-derived Viral Production Cells 2.0 in Viral Production Medium. Optimal viral titers are mediated by our advanced lipid nanoparticle transfection reagent in combination with a novel production enhancer. The components are designed to work synergistically, resulting in maximal titers and eliminating the need to optimize reagents and protocols.

Features of the AAV-MAX Helper-Free AAV Production System include:

  • Components work synergistically to produce high AAV titers greater than 5 x 1010 vg/mL (unpurified)
  • Suspension-based system has scalable protocols from shake flask to bioreactor scale

Can I use other transfection reagents, such as PEI (polyethylenimine), in the AAV-MAX Helper-Free AAV Production System?

The AAV-MAX Transfection Reagent, AAV-MAX Tranfection Booster, and AAV-MAX Enhancer are specifically designed to work synergistically with the Viral Production Medium and Viral Production Cells 2.0 to provide maximum titers. Mixing and matching of reagents is not recommended, as this will decrease the overall performance.

How scalable is the AAV-MAX transfection protocol?

The AAV-MAX Enhancer is important for boosting AAV production. If the enhancer is not used, it will reduce system performance by 50% or more.
