How big do DNA fragment homology arms need to be when using Gibson Assembly kits?

For fragments that are 8 kb or lesser in length, an overlap of 20-40 bp is recommended. For fragments greater than 8 kb in length, an overlap of 40-100 bp is recommended.

What type of vector can I use with Gibson Assembly kits?

Any linearized vector can be used with Gibson Assembly kits. We recommend using high-copy vectors for small inserts (<10 kb) and low-copy vectors for large inserts or toxic genes.
