Description
The MP5-20F3 antibody reacts with mouse interleukin-6 (IL-6). The MP5-20F3 antibody is a neutralizing antibody. Mouse IL-6 (also known as B Cell Stimulating Factor, BCSF, and BSF-2) is a 21-28 kDa factor produced by a variety of cell types, including monocytes, fibroblasts and endothelial cells. Upon stimulation, macrophages, T, B, mast, and glial cells, eosinophils, keratinocytes and granulocytes also secrete IL-6. This pleiotrophic cytokine influences the antigen-specific immune response, inflammatory response, and the development of neurons, and is a major mediator of the acute phase reaction.

Applications Tested
This MP5-20F3 antibody has been tested by intracellular staining followed by flow cytometric analysis of mouse thioglycolate-elicited peritoneal exudate cells using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) and protocol. Please refer to "Staining Intracellular Antigens for Flow Cytometry, Protocol A: Two step protocol for intracellular (cytoplasmic) proteins" located at Flow Protocols. This may be used at less than or equal to 0.5 µg per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.

Blocking Buffers
When using two or more Super Bright, Brilliant Violet™, Brilliant Ultra Violet™, or other polymer dye-conjugated antibodies in a staining panel, it is recommended to use Super Bright Complete Staining Buffer (Product # SB-4401) or Brilliant Stain Buffer (Product # 00-4409-75) to minimize any non-specific polymer interactions. Please refer to the datasheet for Super Bright Staining Buffer or Brilliant Stain Buffer for more information.

Light sensitivity
This tandem dye is sensitive to photo-induced oxidation. Please protect this vial and stained samples from light.

Fixation
• Samples can be stored in IC Fixation Buffer (Product # 00-8222) (100 µL of cell sample + 100 µL of IC Fixation Buffer) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency/compensation.
• Some generalizations regarding fluorophore performance after fixation can be made, but clone specific performance should be determined empirically.
• Our internal testing suggests that Brilliant Violet™ 650 (BV650) is not compatible with methanol-based fixation.

Target Information

IL-6 is a gene that encodes a cytokine that plays a role in inflammation and the maturation of B cells. The protein produced by this gene is an endogenous pyrogen that can induce fever in people with autoimmune diseases or infections. The gene is primarily expressed at sites of acute and chronic inflammation, where it is secreted into the serum and induces a transcriptional inflammatory response through interleukin 6 receptor, alpha. The functioning of this gene is implicated in a wide variety of inflammation-associated disease states, including suspectibility to diabetes mellitus and systemic juvenile rheumatoid arthritis. Elevated levels of the encoded protein have been found in virus infections, including COVID-19 (disease caused by SARS-CoV-2). Diseases associated with IL6 include Kaposi Sarcoma and Rheumatoid Arthritis, Systemic Juvenile.
