Description

Two SeCore DRB sequencing strategies are available to best suit your resolution and throughput needs; DRB1 Locus Sequencing Kit (Single Amplification System) CE and DRB1, DRB3, 4 and 5 Sequencing Kit CE (Group Specific Set). Both formats are designed for bi-directional sequencing of exon 2 and include a Codon 86 Sequencing Mix.

Strategy 1:

The SeCore DRB1 Locus Sequencing Kit CE (Single Amplification System) begins with only one PCR per sample. This method eliminates the need for a confirmatory agarose gel and offers the user the most high throughput route for DRB1 sequencing.

Strategy 2:

The SeCore DRB1, DRB3, 4 and 5 Sequencing Kit CE (Group Specific Set) provides eleven group specific amplification mixes. All eleven amplification mixes can be used for each sample, or the user can select only the amplification mixes necessary for each sample based on previous low-resolution data. The resulting typing data for each separated allele is a greatly simplified homozygous electropherogram, eliminating the challenge of interpreting polymorphic positions. This strategy reduces the cis/trans ambiguities for DRB locus by 98%.

Additional Resolution: SeCore Group Specific Sequencing Products (GSSP) can further resolve DRB1 ambiguities. The amplicon from either SeCore DRB core kits serves as the template. GSS-Products contain a single group specific unidirectional sequencing mix and precipitation buffer. Sequencing parameters are identical to the other core SeCore products. All GSS-Products are sold Research Use Only (RUO).

The SeCore DQB1 Locus Sequencing Kit CE amplifies bidirectionally sequences exon 2 and exon 3.

The SeCore DPB1 Locus Sequencing Kit CE includes four amplification mixes, each specific to a set of alleles. Heterozygote sample alleles are often amplified by different mixes and therefore are separated by this initial PCR. The resulting typing data for each separated allele is a greatly simplified electropherogram, eliminating the challenge of interpreting polymorphic positions. This strategy reduces the cis/trans DPB1 Locus ambiguities by 70%.

The SeCore® HLA Sequencing System is intended for the identification and definition of Class I and II Human Leukocyte Antigens (HLA) and is for use in HLA typing. The SeCore® HLA Sequencing System provides human histocompatibility information of HLA Class I (A, B, and C) and Class II (DPB1, DQB1, and DR) loci using genomic DNA isolated from whole blood specimens. HLA typing using the SeCore® Sequencing Kits must be performed in the presence of an HLA Lab Director, Technical Supervisor and/or general Supervisor following accepted laboratory accreditation standards (ASHI). These products are for professional use only. This test must not be used as the sole basis for making clinical decisions.
