The 1x eBioscience TMB Solution is a sensitive chromogenic substrate supplied as single ready-to-use solution. TMB (3,3’,5,5’-Tetramethylbenzidine) is a substrate for horseradish peroxidase (HRP) and is recommended for Ready-SET-Go! ELISA kits.

During the enzymatic reaction the TMB substrate will result in a blue color that can be read on a spectrophotometer at 650 nm. This kinetic reaction can be stopped using 1M phosphoric acid resulting in a color change to yellow that can be read at 450 nm.

常見問答集 (常見問題)

Are all your TMB substrates transparent and do they all turn blue after reacting with the HRP enzyme of an antibody?

No, we offer different TMB substrates for different applications.
We recommend the following TMB substrates for this particular application:
- eBioscience TMB Solution (1X) (Cat. No. 00-4201-56)
- (For higher sensitivity) 1-Step Ultra TMB-ELISA Substrate Solution (Cat. No. 34029)

What are the differences between TMB and Super AquaBlue?

TMB is a faster developing, stronger substrate that yields greater amplification/sensitivity/background. The use of an acid stop solution produces a yellow end product read at 450nm. Super AquaBlue is a slower developing blue/green substrate particularly useful for kinetic studies.

What other reagents will I need to run an ELISA using the antibody pairs?

When working with our matched pairs or uncoated ELISA kits, please refer to the user guide provided with your antibody-matched pair for kit-specific guidance. When designing your own ELISA, please utilize our ELISA Builder tool for reagent selection as well as our ELISA Development and Optimization pages for futher guidance.
