AGO1 Antibody (711755) in WB

Western blot analysis was performed on whole cell extracts (30 ug lysate) of MCF-7 (Lane 1), NTERA-2 cl.D1 (Lane 2), HEL 92.1.7 (Lane 3), K-562 (Lane 4), MOLT4 (Lane 5), A-431 (Lane 6), THP-1 (Lane 7), MDA-MB-231 (Lane 8) and T-47D (Lane 9). The blots were probed with ABfinity™ Anti-AGO1 Rabbit Oligoclonal Antibody (Product ## 711755, 1-2 ug/ml). A 97 kDa band corresponding to AGO1 was observed across most of the cell lines tested. The blots were detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product ## A27036, 0.4 ug/ml, 1:5000 dilution). Known quantity of protein samples were electrophoresed using Novex® NuPAGE® 4-12% Bis-Tris gel (Product ## NP0321BOX), XCell SureLock™ Electrophoresis System (Product ## EI0002) and Novex® Sharp Pre-Stained Protein Standard  (Product ## LC5800). Resolved proteins were then transferred onto a nitrocellulose membrane with iBlot® Dry Blotting System (Product ## IB21001). The membrane was probed with the relevant primary and secondary Antibody following blocking with 5% skimmed milk. Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product ## WP20005).
