Description: This rabbit polyclonal antibody reacts with mouse and rat Receptor Interacting Protein (RIP) 3. This 57 kDa Ser/Thr kinase is ubiquitously expressed and resides primarily within the cytoplasm. However, studies have suggested that this protein can shuttle between the nucleus and cytoplasm. Although its kinase domain is similar to that of RIP1 and 2, RIP3 does not possess a death domain or CARD motif. RIP3 binds to and phosphorylates RIP1, resulting in inhibition of RIP1- and TNFR1-mediated NFkB activation. RIP3 mediates programmed necrosis by regulating TNF-induced reactive oxygen species production. Moreover, studies have also identified RIP3 as the molecular switch between TNF-induced apoptosis and necrosis in some cell types.
Applications Reported: This polyclonal antibody has been reported for use in western blotting and immunohistochemical staining.
Applications Tested: This polyclonal antibody has been tested by western blot analysis of NIH-3T3 cell lysate. This can be used at 0.5-4 µg/mL. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
Purity: Greater than 90%, as determined by SDS-PAGE.
Aggregation: Less than 10%, as determined by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.
RIPK3 is a member of the receptor-interacting protein (RIP) family of serine/threonine protein kinases, which contains a C-terminal domain unique from other RIP family members. RIPK3 is predominantly localized to the cytoplasm, and specifically, are colocalized in the mitochondrion which can undergo nucleocytoplasmic shuttling dependent on novel nuclear localization and export signals. RIPK3 is a component of the tumor necrosis factor (TNF) receptor-I signaling complex, and can induce apoptosis and weakly activate the NF-kappaB transcription factor and caspase-8 prevents RIPK3-dependent necrosis without inducing apoptosis by functioning in a proteolytically active complex with CFLAR and that this complex is required for the protective function.
