Description: This R3-5G4 monoclonal antibody recognizes human Runx3, a 44 kDa transcription factor that belongs to the Runt domain-containing family. Runx3 is expressed in hematopoietic cells such as T cells, B cells, and myeloid cells. This transcription factor is also expressed in non-hematopoietic tissues, including the epidermis, bone, and dorsal root ganglia. Runx3 has been established to play a critical role in thymocyte development. More specifically, Runx3 promotes CD8 + T cell development by silencing Cd4 expression. Finally, studies have demonstrated the tumor suppressive activity of in a variety of cancers (e.g., gastric and breast cancers).
Applications Reported: This R3-5G4 antibody has been reported for use in western blotting, and immunohistochemical staining of formalin-fixed paraffin embedded tissue sections.
Applications Tested: This R3-5G4 antibody has been tested by western blotting of the Raji cell line and by immunohistochemistry of formalin-fixed paraffin embedded human skin tissue using the IHC Antigen Retrieval Solution - High pH (cat 00-4956). This can be used at less than or equal to 5 µg per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
Purity: Greater than 90%, as determined by SDS-PAGE.
Aggregation: Less than 10%, as determined by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.
This gene encodes a member of the runt domain-containing family of transcription factors. A heterodimer of this protein and a beta subunit forms a complex that binds to the core DNA sequence 5'-PYGPYGGT-3' found in a number of enhancers and promoters, and can either activate or suppress transcription. It also interacts with other transcription factors. It functions as a tumor suppressor, and the gene is frequently deleted or transcriptionally silenced in cancer. Multiple transcript variants encoding different isoforms have been found for this gene.
