Immunohistochemistry was performed on 4% paraformaldehyde-fixed paraffin-embedded mouse brain tissue sections. To expose target antigens, tissues were treated with 15ug/ml Proteinase K in PBS containing 0.01% Triton X-100 for 5 minutes. Tissues were blocked for 1 hour and probed with an Agouti Related Protein polyclonal antibody (Product # PA1-18414) at a dilution of 1:200 overnight at 4°C. Tissues were washed extensively in TBST. Detection was performed using a fluorescently-conjugated goat anti-guinea pig IgG secondary antibody (red) at a dilution of 1:200 for 1 hour at room temperature.
