CtBP2 Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemistry was performed on paraffin-embedded adenocarcinoma of human colon tissue. To expose target proteins, heat-induced epitope retrieval by 1mM EDTA in 10mM Tris buffer (pH8.5) at 120°C for 3 min. Following antigen retrieval, tissues were probed with a CTBP2 monoclonal antibody (Product # MA5-26926) at a dilution of 1:150.

CtBP2 Antibody in Western Blot (WB) — Western blot was performed using anti-CtBP2 Monoclonal Antibody (Product # MA5-26926) a ~48 kDa band corresponding to CtBP2 was observed across the cell lines tested except Raji which is reported to be negative. Modified Whole cell extracts of (30ug lysate) (1% SDS) of HeLa (Lane 1), OVCAR-3 (Lane 2), MOLT-4 (Lane 3) and Raji (Lane 4) were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001).The blot was probed with the primary antibody (1:2000 dilution) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

CtBP2 Antibody — Antibody specificity was demonstrated by detection of differential basal expression of the target across cell lines tested owing to their inherent genetic constitution. Relative expression of CtBP2 was observed in HeLa, OVCAR-3 and MOLT-4 in comparison to Raji using Anti-CtBP2 monoclonal antibody (Product # MA5-26926) in Western Blot. {RE}

Target Information

This gene produces alternative transcripts encoding two distinct proteins. One protein is a transcriptional repressor, while the other isoform is a major component of specialized synapses known as synaptic ribbons. Both proteins contain a NAD+ binding domain similar to NAD+-dependent 2-hydroxyacid dehydrogenases. A portion of the 3' untranslated region was used to map this gene to chromosome 21q21. 3; however, it was noted that similar loci elsewhere in the genome are likely. Blast analysis shows that this gene is present on chromosome 10.
