Sandwich ELISA analysis using a ten point standard curve plotted with a full-length active recombinant EGF protein standard at concentrations ranging from 12.5 ng/mL to 0.3 pg/mL bound by anti-EGF capture antibody coated onto the plate. The standard was detected using a biotin-conjugated EGF recombinant rabbit oligoclonal antibody (Product # 710569) at a concentration of 2ug/mL. A non-linear regression analysis was performed (4 PL) and LOD and LOQ for the antibody pair was determined.

Epidermal growth factor (EGF) is a growth factor which belongs to the protein kinase superfamily. EGF is far and wide expressed in kidney, cerebrum, prostrate and salivary glands. When EGF binds to its receptor EGFR; it stimulates cell growth, proliferation and differentiation. Encoded protein acts as a potent mitogenic factor that plays several important roles. The phosphorylated receptor recruits adapter proteins like GRB2 which in turn activates complex downstream signaling cascades. EGF activates at least 4 major downstream signaling cascades including the RAS-RAF-MEK-ERK, PI3 kinase-AKT, PLCgamma-PKC and STAT modules. Studies suggest the protein may also play important rol ein activating the NF-kappa-B signaling cascade.
