This antibody is predicted to react with porcine based on sequence homology.
Heat-mediated antigen retrieval is recommended prior to staining, using a 10mM citrate buffer, pH 6.0, for 10 minutes followed by cooling at room temperature for 20 min. Following antigen retrieval, incubate samples with primary antibody for 30 min at room temperature. A suggested positive control is breast carcinoma.
The human estrogen receptor (ER) exists in two forms, ERa and ERß. The receptors have a high degree of homology in their DNA binding and ligand binding domains and can form heterodimers. However, the two ERs signal in different ways; for example, ERß does not potentiate the nuclear transcription factor AP-1 when complexed with oestradiol, whereas ERa does. This suggests that ERa and ERß may have different roles in gene regulation and their relative levels within tissues may influence cellular responses to oestrogens.
