This product is a cocktail containing mouse monoclonal antibody MA5-12117 (kappa) and rabbit polyclonal PA5-16651 (lambda). Kappa-lambda double staining contributes to the observation of clonality of the infiltrate. This product is provided at an optimal concentration in an easy-to-use format for demonstrating clonality. A ready-to-use MultiVision polymer detection system consisting of an anti-mouse (HRP) + anti-rabbit (AP) is available (Thermo Scientific Product # TL-012-MHRA). This cocktail is specially formulated from MultiVision polymers that provide increased sensitivity, time-savings and detection simplicity. The MultiVision AP and HRP polymers are innovative, patented technology. The small amino acid polymer subunits minimize conflicts in binding the target protein resulting in more consistent staining and better signal amplification. Ultimately, this gives the user higher sensitivity and antibody efficiency as well as better signal-to-noise ratios. This system is biotin-free, which prevents background staining found with traditional biotin-based methods. MultiVision polymer detection system is a simple and robust one-step polymer detection system for double staining of kappa light chain and lambda light chain in red and blue, respectively. The MultiVision polymer detection system yields high quality double staining in less than 2 hours on formalin-fixed and paraffin-embedded tissue sections. The kit allows observation of two single-stained cells in blue (lambda light chain) and red (kappa light chain) and/or co-localization marked by a purple/brownish mixed-color. This MultiVision Kappa -Lambda cocktail along with the MultiVision polymer detection system has also proven to work with formalin-fixed and BMA+MMA (1:1) embedded plastic tissue sections.
The antibodies to lambda and kappa light chain are reported as useful in the identification of leukemias, plasmacytomas and certain non-Hodgkin lymphomas. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is clonal and therefore malignant.
