Immunohistochemistry analysis of formalin-fixed, paraffin-embedded human brain tissue stained using a Microtubule-associated proteins 1A/1B light chain 3A precursor (MAP1 LC3A) polyclonal antibody (Product # PA5-23166) at 10 ug/ml. Staining of formalin-fixed tissues is enhanced by boiling tissue sections in 10 mM sodium citrate buffer, pH 6.0 for 10-20 min followed by cooling at RT for 20 min.

MAP1 LC3A is a structural protein and a component of both MAP1A and MAP1B microtubule-binding complexes which mediate the physical interactions between microtubules and components of the cytoskeleton. A member of the MAP1LC3 family, LC3A is known to have an ubiquitin fold at the C-terminal region and 2 helices at the N-terminal region. The posttranslational modification of LC3A precursor molecule leads to cleavage by APG4B/ATG4B, resulting in the cytosolic form, LC3-I which is further activated and modified to form the membrane-bound form, LC3-II. LC3A, a mammalian homolog of yeast Atg8, plays an essential role in autophagy, leading to bulk degradation of cytoplasmic components by the lysosomal system where it acts as an adaptor protein between microtubules and autophagosomes. Abundant expression of LC3A is seen in heart, brain, liver, skeletal muscle and testis but absent in thymus and peripheral blood leukocytes.
