PA3-101 detects the glycine-binding protein.
PA3-101 has been used successfully in Western blot ELISA, immunohistochemistry, immunocytochemistry, and immunoprecipitation procedures. In Western blot analysis of rat brain synaptic membranes this antibody detects a ~60 kDa protein representing glycine-binding protein.
The PA3-101 immunogen is a synthetic peptide corresponding to the glycine-binding region residues C(311) G E R N E S F T E E G L D L H(326) of rat GBP.
Ionotropic glutamate receptors are oligomeric ion channel proteins, which mediate fast neurotransmission and excitatory synapses in the mammalian central nervous system. There are three distinct subfamilies, of which NMDA receptors have received particular attention because they are implicated in both synaptic plasticity and the pathogenesis of acute and chronic neurodegenerative disorders. A unique feature of the NMDA receptors is their requirement for both glutamate and the co-agonist glycine for efficient gating. Glycine increases the affinity of the receptor for the endogenous agonist glutamate. The glycine binding site is found on the NR1 subunit of the NMDA receptor.
