Heat-mediated antigen retrieval is recommended prior to staining, using a 1mM EDTA buffer, pH 8.0, for 10 minutes followed by cooling at room temperature for 20 min. Following antigen retrieval, incubate samples with primary antibody for 10 min at room temperature. A suggested positive control is prostate adenocarcinoma with XMRV infection.
Xenotropic murine leukemia virus-related virus (XMRV) is a newly recognized human retrovirus first identified in prostate cancer tissues from men with a deficiency in the innate immunity gene RNASEL. At present, studies have detected XMRV DNA and protein in prostate cancer at widely different rates and in patients with chronic fatigue syndrome. The XMRV protein is synthesized in the Golgi by host cells.
