Bcl-2 (B-cell lymphoma) and close homologues promote cell survival while many of its relatives lead to apoptotic cell death. The Bcl-2 homology 3 (BH3) domain is a potent death domain, and a novel BH3 domain containing protein, termed Bim or BOD (Bcl-2-related Ovarian Death Gene), has recently been cloned. Bim possesses a hydrophobic C-terminus and is localized to the intracytoplasmic membranes. Bcl-2 binds to Bim and modulates its death function, while Bcl-2 mutants are unable to do either.
