Western blot analysis was performed on whole cell extracts (30 ug lysate) of K562 (Lane 1), PC-3 (Lane 2) and Mouse Small Intestine (Lane 3). The blots were probed with ABfinity™ Rabbit Monoclonal Anti-RUNX1 Antibody (Product# 701202, 1-3 ug/ml) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.4 ug/ml, 1:2500 dilution). A ~ 60 kDa corresponding to RUNX1 was observed in K562 and PC3, while ~ 48 kDa band was observed in mouse small intestine tissue lysate. Known quantity of protein samples were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0321BOX), XCell SureLock™ Electrophoresis System (Product # EI0002) and Novex® Sharp Pre-Stained Protein Standard (Product # LC5800). Resolved proteins were then transferred onto a transferred onto a nitrocellulose membrane with Pierce™ Power Blotter System (22834). The membrane was probed with the relevant primary and secondary Antibody following blocking with 5 % skimmed milk. Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

Runx1 belongs to the mammalian RUNX family of DNA binding proteins that regulate the expression of genes involved in cellular differentiation and cell cycle progression. Runx1 is essential for hematopoietic stem cell generation in the vascular regions of the aorta, vitelline and umbilical arteries, yolk sac and placenta. Runx1 is expressed in all tissues except brain and heart, with highest levels in thymus, bone marrow and peripheral blood.
