Ricinus communis agglutinin 60 is a heterodimeric glycoprotein consisting of an A chain (RTA, ricin toxin A chain) disulphide-linked to a B chain (RTB, ricin toxin B chain). RTA is an N glycosidase, responsible for ricin cytotoxicity, that depurinates adenine residue 4324 of the 28S rRNA, thus inactivating protein synthesis. Since its endocytotic properties are very limited, RTA alone shows a very low level of cytotoxicity. RTB is a galactose specific lectin containing three galactose-binding sites. Mutation or deletion of any one of these sites reduces the ability of ricin to penetrate into the cell.
