Heat-mediated antigen retrieval is recommended prior to staining, using a 10mM citrate buffer, pH 6.0, for 10 minutes followed by cooling at room temperature for 20 min. Following antigen retrieval, incubate samples with primary antibody for 10 min at room temperature. A suggested positive control is breast carcinoma.
ERK-1/extracellular signal-regulated kinase 1/p44 MAP kinase/microtubule-associated protein 2 kinase/myelin basic protein kinase/EGF receptor T669 kinase is ubiquitously expressed in various tissues and cells. It gets activated by phosphorylation of neighboring serine and threonine residues by MEK-1 and MEK-2 in response to diverse stimuli like growth factors and mitogens.
