Why are my PCR tubes crushed/deformed after my PCR run?

The heated thermal cycler lid is designed to make sure that optimized pressure is applied on the microplate for an efficient PCR reaction. When using PCR tubes, excess pressure on the tubes, from the heated lid can cause deformation of the tubes. To avoid this, we recommend using the Tray/Retainer set, as shown below:

Cat. No. 4381850, MicroAmp 96-Well Tray/Retainer Set for Veriti Systems – This Tray is for use with MicroAmp Strip Tubes (or MicroAmp Reaction Tubes without Caps, 0.2 mL), with the Veriti Thermal Cycler, SimpliAmp Thermal Cycler, and Proflex PCR System.
Cat. No. 403081, MicroAmp 96-Well Tray/Retainer Set – This Tray is for use with MicroAmp Strip Tubes (or MicroAmp Reaction Tubes without Caps, 0.2 mL), with the 7000 System, 2720 Thermal Cycler, and GeneAmp PCR System 9700.
Cat. No. 4379983, MicroAmp 96-Well Tray for VeriFlex Blocks – This Tray is for use with MicroAmp Strip Tubes (or MicroAmp Reaction Tubes with Caps, 0.1 mL), with the Veriti Thermal Cycler, Veriti Fast Thermal Cycler, SimpliAmp Thermal Cycler, and Proflex PCR System.

My tubes are melting. What is wrong?

This can happen if the wrong plastics are used, or if the tubes are not being used with the correct tray/retainer set. Check the manual for directions on appropriate loading of the sample. Also, you can check for compatible plastics from here (https://www.thermofisher.com/us/en/home/life-science/pcr/pcr-plastics.html).

How do I perform the temperature verification on the 2720 Thermal Cycler?

Please refer to the guidelines in this protocol (https://tools.thermofisher.com/content/sfs/manuals/cms_070449.pdf) specific to the 2720 Thermal Cycler.

How do I program an infinite hold in the 2720 Thermal Cycler?

To add an infinite hold, set the time for the hold to ‘99.59'. The screen should then default to the infinity symbol for your hold.

Is the 2720 Thermal Cycler capable of running a gradient?

No, the 2720 does not have a gradient feature. Our newer thermal cyclers including the ProFlex PCR System and Veriti Thermal Cycler have better-than-gradient VeriFlex blocks.

What is the Auto Delta (AutoX) feature on the 2720 Thermal Cycler?

The AutoDelta feature allows you to incrementally increase or decrease the temperature or hold time with each cycle for a given step within a cycling stage. One common application for this is touchdown PCR.

What is the max block ramp rate of the 2720 Thermal Cycler?

The max block ramp rate of the 2720 Thermal Cycler is 2.3 degrees C/sec.

What types of plates or tubes can I use with the 2720 Thermal Cycler?

Please see the list here (https://www.thermofisher.com/us/en/home/life-science/pcr/pcr-plastics/thermal-cycler-plastics/2720-plastics.html) for compatible plates and tubes.

What types of blocks are available on the 2720 Thermal Cycler?

The 2720 Thermal Cycler contains a 96-well standard block, which can be used with 0.2 mL tubes and plates.

Which thermal cyclers have been tested for use with Cytoscan HD?

We recommend using the following thermal cyclers with Cytoscan HD:

For pre-PCR, we recommend 2720 Thermal Cycler (Cat. No. 4359659)

For pre- and post-PCR , we recommend the following thermal cyclers:
- GeneAmp PCR System 9700 Gold (Cat. No. 4314878) (Note: This instrument has been discontinued and therefore cannot be ordered)
- GeneAmp PCR System 9700 Silver (Cat. No. N8050001) (Note: This instrument has been discontinued and therefore cannot be ordered)
- Applied Biosystems Veriti Dx 96-Well Thermal Cycler (Cat. No. 4452300)
- Applied Biosystems Veriti 96-Well Thermal Cycler (Cat. No. 4375786)
- Bio-Rad® DNA Engine® PTC-200 Thermal Cycler
- Eppendorf® Mastercycler® Pro S Thermal Cycler

Note: You must use thermal cyclers with silver or gold-plated silver blocks. Do not use thermal cyclers with aluminum blocks.
