Why are my PCR tubes crushed/deformed after my PCR run?

The heated thermal cycler lid is designed to make sure that optimized pressure is applied on the microplate for an efficient PCR reaction. When using PCR tubes, excess pressure on the tubes, from the heated lid can cause deformation of the tubes. To avoid this, we recommend using the Tray/Retainer set, as shown below:

What should I do if my Veriti Thermal Cycler fails the temperature non-uniformity test?

If the test fails:

1.Verify that the:
-Calibration of the TVK is up-to-date.
-Probe tips are not cracked or damaged.
2.Dispense 2 to 3 drops of mineral oil onto a cotton swab. Use the swab to apply a thin film of oil to the cone surface of each probe to improve heat transfer.
3.Repeat the steps in “Before You Run 96-Well Instrument Tests” from the Multiprobe Temperature Verification Kit User Guide (http://tools.thermofisher.com/content/sfs/manuals/4460108A.pdf).
4.Verify that the:
-Probes are seated securely in the appropriate test wells.
-Cables are connected securely to the ports.
-Ribbon cable lies flat under the thermal cutout and is not crimped.
-Heated cover is closed and locked down.
5.Repeat the steps in “Run the Test” from the Multiprobe Temperature Verification Kit User Guide (http://tools.thermofisher.com/content/sfs/manuals/4460108A.pdf). Please note, if the test fails again, contact us at instrumentservices@thermofisher.com.

My tubes are melting. What is wrong?

This can happen if the wrong plastics are used, or if the tubes are not being used with the correct tray/retainer set. Check the manual for directions on appropriate loading of the sample. How do I program an infinite hold in the Veriti Thermal Cycler?

To add an infinite hold, select the infinity symbol for program.

How do the VeriFlex Blocks work on the Veriti Thermal Cycler?

The Veriti Thermal Cycler with a 96-well block (Fast and Standard) features innovative VeriFlex Blocks for enhanced PCR functionality. Six separate Peltier blocks provide maximal versatility and flexibility and offer two key benefits:
1. PCR optimization: each block can be set with a specific temperature, which is ideal for precise control over PCR optimization.
2. Run more experiments: the six Peltier blocks can also be used to run up to six different annealing temperatures in the same run.

What is the Auto Delta feature on the Veriti Thermal Cycler?

The AutoDelta feature allows you to incrementally increase or decrease the temperature or hold time with each cycle for a given step within a cycling stage. One common application for this is touchdown PCR.

What is the max block ramp rate of the Veriti Thermal Cycler?

The 96-Well Block has a 3.9 degrees C/sec max block ramp rate, the 96-Well Fast Block has a 5.0 degress C/sec max block ramp rate, the 384- Well Block has a 3.7 degrees C/sec max block ramp rate, and the 60-Well Block has a 3.3 degrees C/sec max block ramp rate.

What types of plastics can I use with the Veriti Thermal Cycler?

The plates and plastics will be specific to the block type. Choose the appropriate block type below for a list of the compatible tubes, plates, and films.

-Veriti 60-Well
-Veriti 96-Well 0.1 mL
-Veriti 96-Well 0.2 mL

What types of blocks are available on the Veriti Thermal Cycler?

The Veriti Thermal Cycler is available in four formats:
Veriti 96-Well Thermal Cycler (a 0.2 mL or 0.1 mL)
Veriti 384-Well Thermal Cycler
Veriti 60-Well Thermal Cycler Veriti 96-Well Fast Thermal Cycler

Have you tested the Veriti and VeritiPro Thermal Cyclers in low humidity conditions (below 20%)?

The recommended conditions are determined to ensure no static discharge (low humidity) occurs.
Recommended Conditions:
- Veriti: 20-80% humidity
- VeritiPro: 15-80% humidity
We have not tested Veriti and VeritiPro thermal cyclers in low humidity, below 20%, therefore we cannot recommend using them in those conditions.

What is the optimal ramp rate for my thermal cycler?

There is no universal ramp rate for a thermal cycler because both the block and sample ramp rates are different for each thermal cycler. The sample ramp rate changes with the volume of the sample since more time and energy are required to heat higher sample volumes. The ramp rate may also need to be changed for a particular PCR assay or primer pair if you are trying to use the same PCR assay on two different thermal cyclers, for example.

As a reference, the block ramp rate commonly used with DreamTaq Polymerase on the MiniAmp thermal cycler is 3.0 C/sec. In a 50 µL PCR reaction, this translates to a sample ramp rate of ~1 C/sec.
