At what temperature should my Mr. Frosty controlled rate cell freezing container be when I start freezing cells?

For best cooling rate performance, the entire system should be at room temperature just prior to freezing.

Should I retain or discard the foam insert in my Mr. Frosty controlled rate cell freezing container?

The white foam insert serves no purpose other than to make the printed instructions on the jar easier to read. Its presence or absence will not affect the cooling rate, and so it can be discarded.

The Mr. Frosty system depends on the specific heat capacity of alcohol in order to provide the optimum cooling rate of 1 degree Celsius per minute. After several cooling and warming cycles, enough vapor condensation has taken place that much of the alcohol in the reservoir has been replaced by water, which affects the heat capacity of the assembled unit such that the cooling rate may be affected.
