Essential 8 Flex Medium is a xeno-free and feeder-free medium that supports the culture of embryonic stem cells (ESCs) and induced pluripotent stem cells (iPSCs) without the need for weekend feeding. It is provided in a convenient two-component kit (500 mL basal medium and 10 mL supplement).

Essential 8 Flex provides all of the benefits of Essential 8 Medium: minimized variability of components, reliable and robust cultures with a xeno-free, cGMP produced medium, along with the added benefit of flexibility. Maintenance of standard PSCs no longer requires being in the lab seven days a week.

Flexibility

With Essential 8 Flex Medium, pluripotent stem cells (PSCs) need only be fed on the day of splitting and the day after. If the cells will be left without feeding for more than 48 hours, such as over a weekend, then simply double the feed volume on Friday.

Reliable and robust cultures

Essential 8 Flex Medium has been shown to support PSC growth for >30 passages without any signs of karyotypic abnormalities and to maintain the ability of iPSCs to differentiate into all three germ line lineages.

Cost effective

Essential 8 Flex Medium is provided in a convenient two-component kit (500 mL basal medium and 10 mL supplement), and when used with vitronectin (VTN-N), provides a cost effective, defined system for feeder-free culture of human pluripotent stem cells.

The Essential 8 Flex Medium formulation is based upon that published for Essential 8 Medium by Chen, et al., and allows for control and flexibility of PSC culture medium, without a negative impact to the health of the stem cells.

Both Gibco Vitronectin (VTN-N) Recombinant Human Protein, Truncated (Cat. No. A400457 and A400458) and Gibco Geltrex Flex LDEV-Free hESC-qualified Reduced Growth Factor Basement Membrane Matrix (Cat. No. A4000046801, A4000046802, or A4000046803) have been successfully used with Essential 8 Flex Medium.

Can PSCs transitioned over into Essential 8 Medium on recombinant human laminin-521 (rhLaminin-521) from feeder-dependent cultures, then be transferred onto vitronectin (VTN-N)?

Yes. Following 2 passages on the rhLaminin-521 matrix, Versene or EDTA passaging should be used to subculture PSCs.

What are the main differences to be taken into consideration when culturing cells in Gibco Essential 8 Medium on Gibco Vitronectin (VTN-N) compared to other feeder-free systems?

Here are three major differences to be taken into consideration when culturing cells in Gibco Essential 8 Medium on Gibco Vitronectin (VTN-N) compared to other feeder-free systems:
- Cells should be typically passaged ~24 hours sooner than they would be with other feeder-free media.
- Passaging should take place when cells are at ~85% confluency. If cells are passaged when they are more than 85% confluent, the health of the cells and final cell yield may be compromised.
- Cells must be passaged in EDTA. Collagenase and dispase are not recommended.

Can pluripotent stem cells (PSCs) previously cultured in other media and on other substrates be cultured in Gibco Essential 8 Medium and on Gibco Vitronectin?

Cells cultured in other feeder-free media systems, such as mTeSR Medium with Matrigel Basement Membrane Matrix, or StemPro hESC SFM with Geltrex Matrix, can be successfully cultured in Gibco Essential 8 Medium and VTN-N. In addition, PSCs grown on feeders with KnockOut SR have also been shown to be successfully cultured in Gibco Essential 8 Medium on VTN-N. However, when changing media systems, cells must be passaged either manually, or with EDTA prior to culturing in Gibco Essential 8 Medium on VTN-N.

Can I use enzymes such as dispase and collagenase for passaging cells cultured in Gibco Essential 8 Medium and on VTN-N?

Enzymes such as dispase and collagenase do not work well with cells cultured in Gibco Essential 8 Medium on VTN-N. Use of these enzymes for passaging cells results in compromised viability and attachment.
