Luria Broth (LB) is a popular, rich medium used to cultivate bacteria and to perform coliphage plaque assays. The original formulation published in 1951 by Guiseppe Bertani to study phage plaque formation and named Lysogeny Broth. It is also known as Lennox Broth and Luria-Bertani Broth.

Widely used LB medium formulations differ in salt concentration:

  • Miller: 10 g/L NaCl
  • Lennox: 5 g/L NaCl
  • Luria: 0.5 g/L NaCl

Luria Broth Base (Miller's formulation) is recommended for the general maintenance and growth of E. coli as it allows for fast cell expansion and supports high cell density. In cases where salt-sensitive antibiotics are being used for selection (such as Zeocin, tetracycline or chloramphenicol), Lennox formulation medium would be recommended.

Luria Broth Base formulation

Formulation grams per liter: Pancreatic digest of casein 10.0, Yeast extract, 5.0 Sodium chloride 10.0

To prepare:  Suspend 25.0 g in 1 litre of distilled water. Mix well and distribute into final containers. Sterilize by autoclaving at 121°C for 15 minutes.
