Description

The Thermo Scientific Human IL-8 ELISA Kit is used to measure IL-8 (human interleukin-8) levels in human serum, plasma (EDTA, heparin, sodium citrate), urine and culture supernatants. Each kit contains one 96-well Strip Plate precoated with a human IL-8 capture antibody and sufficient detection antibody, conjugate, buffers, diluent, TMB and stop buffers, as well as lyophilized human IL-8 standard to analyze several samples.

Features of the Thermo Scientific Human IL-8 ELISA Kit:

Target: natural and recombinant human interleukin-8 (IL-8) and has been shown to recognize IL-8 derived from stimulated human umbilical vein endothelial cells
Format: colorimetric ELISA (TMB substrate) in clear 96-well strip plate
Measurement: absorbance at 450nm minus absorbance at 550nm
Assay Range: 25.6 to 1000 pg/mL
Sensitivity: < 2 pg/mL
Precision: intra-assay CV < 10%; inter-assay CV < 10%
Sample Types: cell culture supernatant, urine, serum, plasma (EDTA, sodium citrate and heparin)
Total Assay Time: 3 hours
Sample Size: 50 µL
Specificity: no significant cross-reaction with with human PF-4, GROα, IL-1α, IL-1β, IL-2, IL-3, IL-4, IL-6, IL-7, IL-10, IL-12, TNFα, IFNα, IFNγ, or GM-CSF.

Note: Assay range and other values listed above are for the pre-coated plate kit; see product documentation for values specific to the reagent kit and antibody-pair kit.

These human IL-8 ELISA kits use pre-titered, matched pairs of coating and detection antibodies to achieve sensitive and accurate measurement of interleukin-8 (IL-8) in samples at 50 to 100 µL per well. Also included are calibrated recombinant protein standard, streptavidin-HRP, TMB substrate solution and stop solution. The complete kits contain precoated 96-well strip plate(s) and lot-optimized reagents. The economically-priced reagent kit provides validated reagents only, requiring users to coat their own plates. The antibody-pair kit consists of the matched IL-8 antibody pairs and IL-8 standard only. All components undergo quality control testing to minimize lot-to-lot variability.

Includes:
Complete Kits contain pre-coated antibody-plates, detection antibodies, buffers, diluents, standards, and substrates. The Reagent Kit (previously called a "Screening Set") contains matched antibodies, standard and detection reagents but no coated plates. The Antibody Pair Kit (previously called a "MiniKit") contains only matched antibodies and standard (no plates or detection reagents).

Interleukin-8 is a member of the chemokine alpha superfamily of proteins, which contains at least 14 structurally related cytokines. These cytokines exhibit 20-45% homology, are all basic heparin binding proteins and act on various inflammatory cell types. IL-8 binds to specific cell surface receptors causing a rapid rise in cytosolic Ca++ concentration. Apparent biological effects include neutrophil infiltration, T cell infiltration, angiogenesis, synovial inflammation and plasma leakage.
