The Mouse IL-1a ELISA research-use-only kit is to be used for the quantitative determination of IL-1a in samples (e.g., cell culture supernatants, plasma, serum) using 96-well plates and a microplate reader. The assay will recognize both natural and recombinant forms of IL-1a.

Performance characteristics

• Sensitivity: 10 pg/mL
• Standard curve range: 10 pg/mL–3000 pg/mL
• Sample types: cell culture supernatants, plasma, serum
• Specificity: This ELISA kit shows no cross-reactivity with the following cytokines tested: Mouse CD30, L CD30, T CD40, CRG-2, CTACK, CXCL16, Eotaxin , Eotaxin-2, Fas Ligand, Fractalkine, GCSF, IFN- gamma, IGFBP-3, IGFBP-5, IGFBP-6, IL-1 alpha, IL-1 beta, IL-13, KC, Leptin R, LEPTIN(OB), LIX, L-Selectin, Lymphotactin, MCP-5, M-CSF, MIG, MIP- 1alpha, MIP-1 gamma, MIP-2, MIP-3 beta, MIP-3 alpha, PF-4, P-Selectin, SCF, SDF-1 alpha, TARC, TCA-3, TECK, TIMP-1, TNF RI, TPO, VEGF.
• Sample volume: 100 µL per sample after dilution
• Total assay incubation time: 5 hours

Principle of the method
The Thermo Scientific™ Pierce™ Mouse IL-1a ELISA is a solid-phase, sandwich-type enzyme-linked immunosorbent assay (ELISA). An antibody specific for IL-1a has been coated onto the wells of the microtiter strips provided. Samples, including standards of known IL-1a content, control specimens, and unknowns, are pipetted into these wells, followed by the addition of a biotinylated second antibody.

During the first incubation, the IL-1a antigen binds simultaneously to the immobilized (capture) antibody on one site, and to the solution-phase biotinylated antibody on a second site.

After removal of excess second antibody, streptavidin-peroxidase (enzyme) is added. It binds to the biotinylated antibody to complete the four-member sandwich. After a second incubation and washing to remove all unbound enzyme, a substrate solution is added, which is acted upon by the bound enzyme to produce color. The intensity of this colored product is directly proportional to the concentration of IL-1a present in the original specimen.
