The DES® BioEase™ Expression Kit with pCoBlast provides an easy, efficient method for expressing, purifying, and detecting biotinylated recombinant proteins. The pMT/BioEase™-DEST vector incorporates a 72-amino acid sequence from K. pneumoniae that directs in vivo biotinylation of a specific lysine residue. Proteins produced in pMT/BioEase™-DEST are expressed as fusion proteins to this sequence. Biotinylated fusion proteins can be purified on streptavidin agarose or detected on
western blots using streptavidin-HRP or streptavidin-AP conjugates. In addition to the BioEase™ tag, the pMT/BioEase™-DEST vector offers the following features:

attR sites for fast and efficient recombination with any attL-flanked Gateway® vector
metallothionein (MT) promoter for high level, inducible expression in S2 cells
• Enterokinase (EK) cleavage site for efficient removal of the N-terminal BioEase™fusion tag
• A Blasticidin vector (pCoBlast) for rapid selection of stable cells

Contents & storage

The DES® BioEase™ Expression Kit contains 6 µg of the pMT/BioEase™-DEST vector, 10 µg of the pMT/BioEase™-GW/lacZ expression control, and 20 µg of the pCoBlast selection vector. It also contains the Calcium Phosphate Transfection Kit, frozen S2 cells, GIBCO™ Schneiders Drosophila Medium, and 50 mg of Blasticidin. Store S2 cells in liquid nitrogen. Store medium at +4°C. Store all other components at -20°C. All components are guaranteed stable for 6 months when properly stored.
