Description

The Human Interferon-α (IFN-α) Multi-Subtype ELISA research-use-only kit is to be used for the quantitative determination of human IFN-α in cell culture supernatant samples using 96-well plates and a microplate reader. The kit is specifically formulated to detect 14 out of 15 identified human IFN-α subtypes: IFN-αA, IFN-α2, IFN-αD, IFN-αB2, IFN-αC, IFN-αG, IFN-αH, IFN-αI, IFN-αJ1, IFN-αK, IFN-α1, IFN-α4a, IFN-α4b, and IFN-αWA. The assay is based on the international reference standard for human IFN-α provided by the National Institutes of Health* and recognizes both natural and recombinant forms of human IFN-α.

Principle of the method
The Human IFN-α Multi-Subtype kit is a solid phase sandwich enzyme-linked immunosorbent assay (ELISA). A monoclonal antibody specific for human IFN-α has been coated onto the wells of the microtiter strips provided. Samples, including standards of known human IFN-α content, control specimens, and unknowns, are pipetted into these wells. During the first incubation, the human IFN-α antigen binds to the immobilized (capture) antibody on one site. After washing, a detection antibody specific for human IFN-α is added. During the second incubation, this antibody binds to the immobilized human IFN-α captured during the first incubation.

After removal of excess second antibody, an anti-secondary antibody conjugate (horseradish peroxidase) is added. This binds to the detection antibody to complete the four-member sandwich. After a second incubation and washing to remove all the unbound conjugate, a substrate solution is added, which is acted upon by the bound enzyme to produce color. The intensity of this colored product is directly proportional to the concentration of human IFN-α present in the original specimen.

Background information
Interferons (IFNs) are a family of mammalian cytokines initially characterized by their ability to inhibit viral infection. They are synthesized and secreted by most cell types in response to pathogens. In addition to their antiviral properties, IFNs have also been shown to exhibit anti-proliferative, immunomodulatory, and many other activities.

In humans, IFN-α consists of a group of proteins that are greater than 85% homologous by amino acid sequence. Numerous individual human IFN-α subtypes have been identified; many with different properties. It remains unclear why there are multiple IFN-α subtypes. A variety of studies suggested they possess overlapping but also unique sets of biological activities.
