Detect and differentiate Pseudomonas aeruginosa from other Pseudomonas spp. based on fluorescein production using Thermo Scientific™ Remel™ Pseudomonas F Agar. In 1954, King et al. developed two media formulations (Medium A and Medium B) to enhance pigment production in pseudomonads1. Earlier work had demonstrated that the production of pyocyanin and fluorescein is influenced by amino acids, inorganic ions, minerals, and peptones. King determined that the composition of the peptone in the medium affected the production of these pigments.

Pseudomonas F Agar is used for the differentiation of Pseudomonas aeruginosa from other pseudomonads based on fluorescein production. Pseudomonas F Agar was developed by King, Ward, and Raney to give improved fluorescein production. The absence of minimal concentrations of components which may have a detrimental effect influences pigment production. A combination of dipotassium phosphate and magnesium sulfate was found to be most effective in stimulating fluorescein production2.

The medium contains casein and meat peptones in equal amounts, which supply nitrogenous compounds, amino acids, and vitamins necessary for bacterial growth. This ratio of peptones enhances the production of fluorescein and inhibits pyocyanin production. The peptone in the medium contains approximately 1% phosphorus that results in superior fluorescein production. Additives such as dipotassium phosphate and magnesium sulfate enhance fluorescein production. Addition of glycerol to the prepared medium acts as an energy source.
