Differentiate Gardnerella vaginalis from other organisms found in the genitourinary tract with Thermo Scientific™ Remel™ Starch Agar w/Brom Cresol Purple. Starch Agar w/Brom Cresol Purple is a modification of peptone starch dextrose (PSD) agar developed by Smith in 19751. Dextrose is replaced with starch and brom cresol purple is added as an indicator to detect hydrolysis of starch by G. vaginalis.
Gardner and Dukes first isolated G. vaginalis from women with nonspecific vaginitis in 19552. When 224 genital specimens were tested, G. vaginalis was the only organism that grew on Starch Agar w/Brom Cresol Purple and fermented starch, with the exceptions of some strains like streptococci and lactobacilli3. The acid produced by the fermentation of starch causes the pH indicator, brom cresol purple, to change the color of the medium from purple to yellow in the zone surrounding growth.
Starch Agar w/Brom Cresol Purple contains peptone and beef extract that supply nutrients necessary for bacterial growth. Sodium chloride maintains osmotic equilibrium. GCHI Enrichment is a defined supplement which provides vitamins, amino acids, coenzymes and other factors that stimulate growth. Cornstarch neutralizes fatty acids that may be toxic to G. vaginalis and acts as the fermentable substrate in this medium. Brom Cresol Purple is the pH indicator.
