Determine acid production due to carbohydrate fermentation reactions of microorganisms using Thermo Scientific™ Remel™ Heart Infusion Broth with BCP, 1% Adonitol. In 1918, Huntoon prepared the Heart Infusion Broth using beef heart and peptone. Facklam added carbohydrates and brom cresol purple to this broth to help identify streptococci1. Heart Infusion Broth with brom cresol purple, with or without carbohydrates, is recommended for determination of acid production following carbohydrate fermentation reactions of microorganisms, especially streptococci and enterococci2.
Huntoon’s medium containing fresh beef heart and peptone later came to be known as Heart Infusion Broth3. It demonstrated that without the addition of enrichment such as animal blood, the medium could be used to support the growth of nutritionally fastidious microorganisms.
Heart Infusion Broth with BCP contains 10g of Adonitol along with beef heart infusion and casein peptone that supply nitrogenous compounds and amino acids required for the growth of nutritionally fastidious bacteria, including streptococci. Sodium chloride provides essential electrolytes and maintains osmotic equilibrium. Bacterial growth is enhanced by yeast extract which supplies energy as well as essential B vitamins. Adonitol is added to Heart Infusion Broth base in a concentration of 1% and is the carbohydrate source. Brom cresol purple acts as an indicator which changes the color of the medium from purple to yellow on the production of acid after the fermentation of carbohydrate.
