Demonstrate motility in nonfermentative, gram negative bacilli with Thermo Scientific™ Remel™ Motility B Medium w/TTC Indicator.

Motility B Medium was formulated by Gilardi with a reduced agar concentration to facilitate the appearance of motility by non-fermentative, gram-negative bacilli such as pseudomonads 1,2. The Gard plate technique that contains Motility B Medium poured into a petri dish 3 is used to select a motile strain. Kelly and Fulton recommend the addition of 2,3,5-triphenyltetrazolium chloride (TTC) to enhance the macroscopic appearance of motile organisms in motility media4.

Motility B Medium w/TTC Indicator is a semisolid media that is used to demonstrate motility, primarily in nonfermentative, gram negative bacilli. Agar provides a semisolid medium which makes motility interpretations macroscopic. TTC indicator helps to interpret results. Tetrazolium salt is colorless, but as the organism grows the dye is incorporated into the bacterial cells and is reduced to an insoluble red pigment, formazan. This red color forms only in the area where bacteria is growing.

  • Easy to interpret:
    • Positive test- In presence of TTC, red color is observed in the area of growth.
    • Negative test- Growth confined to the stab line. No red color.

Motility B Medium w/TTC Indicator contains casein peptone and yeast extract that supply amino acids, peptides and vitamins to promote growth of nonfermentative gram-negative bacilli. Sodium chloride provide essential electrolytes and maintains osmotic equilibrium. 2,3,5-triphenyltetrazolium chloride (TTC) acts as indicator and helps to interpret results by changing color.
