Differentiate and enumerate microorgansims in urine specimens using Thermo Scientific™ Remel™ CLED Agar (Dehydrated). Mackey and Sandys modified the solid culture medium for the use of with urine cultures first developed in 19601 by substituting lactose and sucrose for mannitol and increasing the concentration of brom thymol blue and agar2. Further addition of cystine and deletion of sucrose resulted in Cystine Lactose Electrolyte Deficient (CLED) Agar which detects bacteriuria by quantitative culture of urine3.

This medium is recommended for use in qualitative procedures for the cultivation of microorganisms from urine specimens. It contains beef extract which supplies carbohydrates and vitamins, casein peptone which supplies amino acids, nitrogenous compounds and peptides necessary for the growth of bacteria. Cystine enhances the growth of cystine-dependant coliforms. Brom thymol blue is a pH indicator which differentiates lactose fermenters producing yellow colonies from lactose nonfermenters. Electrolytes are reduced in order to restrict the swarming of Proteus.
