Cultivate and isolate anaerobic microorganisms from clinical specimens using Thermo Scientific™ Remel™ Schaedler Agar (Dehydrated). In 1965, Schaedler, Dubos, and Costello formulated Schaedler Base to recover bacteria encountered in the gastrointestinal tract and support the growth of fastidious anaerobic microorganisms1. Mata et al. modified Schaedler's formulation by replacing pancreatic digest of casein with tryptic soy broth and reported excellent recovery of anaerobic and microaerophilic organisms2.
The pigmentation of Prevotella and Porphyromonas spp. is enhanced on the addition of lake blood to the prepared medium. In order to facilitate recovery of anaerobic bacteria from specimens containing commensal microbial flora, selective agents were added.
The medium contains casein, soy and meat peptones that provide the nitrogen, amino acids, and peptides necessary for bacterial growth. Yeast extract is a source of B-complex vitamins and serves as a growth enhancer. Sodium chloride supplies essential electrolytes and maintains osmotic equilibrium. Dextrose provides an energy source. The growth of fastidious microorganisms is stimulated by hemin present in the medium.
