Heat-mediated antigen retrieval is recommended prior to staining, using a 10mM citrate buffer, pH 6.0, for 10 minutes followed by cooling at room temperature for 20 min. Following antigen retrieval, incubate samples with primary antibody for 30 min at room temperature. A suggested positive control is tonsil or B cell lymphoma.
MALT1/Paracaspase (MALT1) was independently identified as a member of the human paracaspase family and an interacting partner of B-cell lymphoma (Uren et al., 2000). MALT1 is a caspase-like protein that contains an N-terminal death domain, two Ig-like domains, and a C-terminal caspase-like domain. It binds to Bcl10 through its Ig-like domains and cooperates with Bcl10 to activate NF-kappaB.
