Resurge CD4 Supplement, without glucose and L-glutamine, is a chemically defined supplement formulation to complement a variety of base media. This supplement was developed using several host cell lines and in various culture systems, including shaker flasks and bench-scale bioreactors. Resurge CD4 Supplement, without glucose and L-glutamine, was developed for use in CHO mammalian culture systems and can be used either as a media supplement or a culture system feed.

B-27™ Supplement, minus insulin (Gibco™)

A variation of the most cited neuronal cell culture supplement, B-27 Supplement Minus Insulin is a customized version of the original B-27 Supplement without insulin. B-27 Supplement Minus Insulin is a serum-free supplement designed to support growth of hippocampal and other CNS neurons in applications where the presence of insulin causes interference. B-27 Supplement Minus Insulin is provided as a 50X liquid and is intended for use with Neurobasal Medium or Neurobasal-A Medium.

B-27 Supplement is suitable for use in most neuronal cell applications. For other applications, we provide alternative formulations of B-27 Supplement and Neurobasal media.

StemPro™ Neural Supplement (Gibco™)

StemPro Neural Supplement is an optimal, convenient, and cost-effective serum-free supplement for the proliferation of mammalian neural stem and progenitor cells such as human and rat neural stem cells (NSC) and glial progenitor cells (GPC). Due to their scarce availability, the in vitro expansion of these cells has been a challenge to academic and clinical researchers. When combined with the appropriate basal medium and growth factors, StemPro Neural Supplement not only supports the expansion of neuronal progenitor cells but also enables the cells to retain their progenitor status and potential to differentiate into neurons and glial cells.

In recent years, there has been an increased scientific interest in studying neural progenitor cells due to their important role in neurogenesis. Recent advances have also pointed to the potential use of neural progenitor cells as effective therapies for neurodegenerative diseases, such as dopaminergic neural progenitor cells for Parkinson’s Disease and oligoprogenitor cells for amyotrophic lateral sclerosis (ALS) and multiple sclerosis (MS).

CTS B-27 Supplement XenoFree is based on the classic B-27 Supplement formulation and contains no animal-derived components at the primary component level. Designed with the translational stem cell researcher in mind, CTS B-27 Supplement XenoFree can be used to support induction of human neural stem cells (hNSCs) from pluripotent stem cells (PSCs), expansion of hNSCs, differentiation of hNSCs, and maintenance of mature differentiated neurons in culture.

Supplement S7 (Gibco™)

Supplement S7 is a sterile, concentrated (100X), ionically balanced solution intended for use as one component in a complete culture environment for human epidermal keratinocytes. Supplement S7 is chemically defined and animal origin-free. Each 5 ml bottle of Supplement S7 is the correct amount of supplement for a 500 ml bottle of EpiLife® basal medium.

Autolyzed Yeast Extract (15% solution) (Gibco™)

Gibco® Autolyzed Yeast Extract, 15% Solution is a cell culture supplement formulated as 150 g Yeast Extract Powder (YEP) per liter of distilled water. YEP is a concentrated extract of soluble amino acids, carbohydrates, vitamins and minerals produced from ultra-filtered Baker's yeast.

Human Keratinocyte Growth Supplement (HKGS) (Gibco™)

A sterile, concentrated (100X) solution intended for use with EpiLife® medium or Medium 154 to culture human epidermal keratinocytes. HKGS is an ionically-balanced supplement containing bovine pituitary extract (BPE), recombinant human insulin-like growth factor-I, hydrocortisone, bovine transferrin, and human epidermal growth factor. Each 5 mL bottle of HKGS contains all of the growth factors, hormones, and tissue extracts necessary for the culture of human epidermal keratinocytes, and is the correct amount of supplement for a 500 mL bottle of medium.

When a 500 mL bottle of medium is supplemented with HKGS, the final concentrations of the components in the supplemented medium are:

• Bovine pituitary extract (BPE), 0.2% v/v
• Recombinant human insulin-like growth factor-I, 1 µg/mL
• Hydrocortisone, 0.18 µg/mL
• Bovine transferrin, 5 µg/mL
• Human epidermal growth factor, 0.2 ng/mL

HAT Supplement (50X)

Gibco® HAT Supplement is a liquid mixture of sodium hypoxanthine (5 mM), aminopterin (20 µM) and thymidine (0.8 mM). HAT-supplemented medium is suitable for post-fusion selection against unfused or self-fused HGPRT–myeloma cells. Hypoxanthine and thymidine supply preformed purines and pyrimidines for DNA synthesis by hybridomas via the salvage pathway that utilizes HGPRT- contributed by the fused spleen cell. Aminopterin, a folic acid antagonist, inhibits the de novo nucleoside biosynthesis pathway.

Resurge CD3 Supplement, without glucose and L-glutamine, is an animal origin-free (AOF) protein-free chemically defined supplement. It can be used in batch and fed batch mode in cell culture processes. All components are chemically defined with known molecular formulae and structures and meet our stringent Animal Origin-Free (AOF) specification.

Resurge CD3 Supplement is formulated without glucose, L-glutamine, and sodium bicarbonate. Due to the balanced nutritional nature of this supplement, rich in amino acids, it has also been used in bacterial applications.

2-Mercaptoethanol (50 mM) (Gibco™)

Gibco® 2-Mercaptoethanol (also known as beta-mercaptoethanol or BME) is a potent reducing agent used in cell culture medium to prevent toxic levels of oxygen radicals. Mercaptoethanol is not stable in solution so most protocols require daily supplementation. Gibco® 2-Mercaptoethanol is made at 50 mM in Dulbecco's Phosphate Buffered Saline (DPBS).

EpiLife™ Defined Growth Supplement (EDGS) (Gibco™)

A defined, sterile, concentrated (100X) solution intended for use with EpiLife® medium to culture human epidermal keratinocytes. EDGS is not intended for use with Medium 154. EDGS contains all of the growth factors and hormones necessary for the growth of human epidermal keratinocytes. Each bottle contains the correct amount of supplement for a 500 ml bottle of EpiLife® medium. EDGS contains:
• purified bovine serum albumin
• purified bovine transferrin
• hydrocortisone
• recombinant human insulin-like growth factor type-1 (rhIGF-1)
• prostaglandin E2 (PGE2)
• recombinant human epidermal growth factor (rhEGF) The bovine products in EDGS have been isolated from animals of North American origin. The isolation process for these components includes steps that inactivate viruses. All components of EDGS are greater than 95% pure.

Resurge CD2 Supplement, without glucose and L-glutamine, was developed for use in CHO mammalian culture systems and can be used either as a media supplement or a culture system feed.

CultureOne™ Supplement (100X) (Gibco™)

CultureOne Supplement is a chemically-defined, serum-free supplement designed to significantly improve the differentiation of neural stem cells (NSCs) to neurons. Compared to conventional differentiation methods where NSCs can overgrow and become burdensome, CultureOne Supplement eliminates more than 75% of contaminating neural progenitor cells with minimal cell death and no effect on other kinase-mediated pathways. The resulting superior neuronal cell cultures of evenly distributed, differentiated neurons enable improved downstream assays, accelerated neuronal maturation, and seamless maintenance for five weeks or more.

Each vial of CultureOne Supplement contains a 100X 5-mL liquid reagent that can be added easily to any conventional neuronal differentiation medium, such as Neurobasal Medium with B-27 Supplements. After two weeks of differentiation with CultureOne Supplement, the supplement can be removed and the resulting cultures will contain considerably fewer progenitor cells for five weeks or more.

Improved downstream assays
Analysis of NSC-derived neurons can be difficult due to the presence of overgrown progenitor cells in the culture. These invasive cells can make imaging and electrophysiology experiments challenging and call into question the physiological relevance of gene expression assays. CultureOne Supplement reduces proliferating progenitor cells, giving you a cleaner and more relevant neuronal cell population, which significantly improves the assay experience and outcome.

Accelerated maturation of differentiated neurons
With CultureOne Supplement, NSC-derived neurons express mature physiological markers faster and develop longer neurites than untreated NSC-derived neuronal cultures. CultureOne Supplement-treated neurons also express significantly higher numbers of voltage gated calcium ion channels, which are an important marker for neural maturity and excitability.

Longer culture of NSC-derived neurons
For experiments that require NSC-derived neurons to mature for long periods of time, CultureOne Supplement seamlessly enables maintenance of enriched, differentiated neurons for five weeks or longer. By contrast, conventional differentiation methods result in proliferating neural progenitor cells, which tend to clump and cause the culture to peel off of the plate.
