Verso 1-Step RT-PCR Kit ReddyMix, with ThermoPrime Taq (Thermo Scientific™)

Thermo Scientific Verso 1-Step RT-PCR Kit ReddyMix (with ThermoPrime Taq) is a complete set of RT-PCR reagents that combines everything needed for RT-PCR. The Verso RT enzyme delivers high processivity and can be used at temperatures up to 57°C, delivering effective transcription through difficult RNA secondary structures. Verso also shows a better dynamic range than standard AMV or M-MuLV enzymes, with increases in template concentration giving rise to corresponding increases in product generated over a wider range of starting material concentrations.

An optional RT Enhancer is provided with the kit and can be used during the RT step to remove any DNA contamination, eliminating the need for DNase I treatment.

This Verso 1-step RT-PCR system comes with the ReddyMix option, where the master mix contains a dye and precipitant that allow RT-PCR reactions to be loaded directly onto an electrophoresis gel. The red dye migrates between bromophenol blue and xylene cyanol at approximately 300 bp, depending on agarose concentration.

TaqPath qPCR Master Mix, CG, is a 2X formulation designed for gene expression and miRNA analysis. It contains thermostable Fast DNA polymerase, uracil-N-glycosylase (UNG), dNTPs with dUTP, ROX dye (passive reference), and optimized buffer components for maximum robustness and reproducibility.

Validated with a breadth of workflows
TaqPath qPCR Master Mix, CG, has been validated to provide high specificity and dynamic range for use in multiple real-time PCR applications. The formulation can be run in either fast or standard cycling conditions on a wide variety of qPCR platforms. The figure below demonstrates the excellent PCR linearity over an 8-log input range of template with both gene expression and miRNA assays.

In addition, TaqPath qPCR Master Mix, CG, has been engineered to retain consistent performance in preassembled reactions for up to 48 hours. The stability of this mix provides users of high-throughput, liquid handling systems the assurance that the results on the first plate will mimic those of the last plate (figure below). Reproducible sensitive detection
We understand the importance of reliably detecting low-copy targets for your test quality and data interpretation. TaqPath qPCR Master Mix, CG, helps generate significant and reproducible CT values for ≤10 copy detection. The figure below shows the consistent CT results obtained from three unique lots when detecting 10-copy inputs. Inhibitor tolerant

Efficiency across a broad range of input concentrations
Maximum flexibility in RNA template input quantity requires optimal real-time PCR efficiency across a broad range of template quantities. Figure 1 shows the excellent amplification efficiency across six orders of magnitude. The result is comparable to that of the TaqMan RNA-to-CT 2-Step Kit, and is a significant improvement in dynamic range and sensitivity over the TaqMan One-Step RT-PCR Master Mix Reagents.

High sensitivity at low target concentration
The sensitivity of TaqMan RNA-to-CT 1-Step Kit was validated using a synthetic RNA template for which copy number is precisely known. Significant sampling error occurs when measuring low quantities of target, so statistical analysis is required for proper evaluation using multiple replicates. Figure 2 shows the expected quantity of target and corresponding mean CT values. Statistical analysis indicates high confidence of sample quantification based on a T-test (Table I), consistent with as few as 10 copies of target*.

*Results are dependent on a variety of factors, including assay design.

The QuantStudio™ 3D Digital PCR 20K Chip Kit v2 is a key component of the QuantStudio™ 3D Digital PCR System. Manufactured from a silicon substrate and etched with 20,000 consistently-sized nanoscale reaction wells, the chip enables precise sample loading with an uncomplicated workflow. It requires use of the QuantStudio 3D Digital PCR Master Mix v2.

QuantStudio 3D Digital PCR Master Mix v2 is specifically formulated with all reagents required to help ensure robust PCR amplification of single copy templates. It is optimized for use with isolated DNA or cDNA, TaqMan™ assays, and the QuantStudio 3D Digital PCR 20K Chip v2.
