Pathatrix™ Auto Instrument Applied Biosystems™

The Pathatrix® Auto Instrument quickly and easily concentrates pathogens and spoilage organisms from a wide range of food and environmental sample matrices for downstream testing by PCR, immunoassay or culture. Because the Pathatrix® Auto Instrument and its companion kits concentrate and isolate specific bacteria such as Salmonella, E. coli O157:H7 and Listeria from sample volumes of up to 60 ml, they support cost-saving pooling protocols, facilitate shorter sample enrichment times to reduce overall time to results, and remove PCR inhibitors that can lead to indeterminate results.

Simple-to-Use, Automated System
The Pathatrix® Auto Instrument automates the concentration and isolation of target bacteria from 10 – 60 ml sample volumes in only about 15 minutes. Simply add a 10-60 ml liquid sample to a sample vessel, add paramagnetic beads that are either antibody-coated or cationic depending on the target organisms of interest, and load the assembly with a filled elution tube into the instrument. Kits for the isolation of Salmonella, E. coli O157:H7, Listeria, Campylobacter and Cronobacter — which include the sample vessel, elution vessel, and corresponding beads — are available separately.

The Pathatrix® Auto Instrument uses a proprietary technology to re-circulate the entire sample over a 'capture phase' where the paramagnetic particles are immobilized. Target organisms are captured onto the immobilized beads, then washed and the beads eluted. The entire process takes only 15 minutes. After elution, the captured organisms, cleaned and purified, are ready for a wide variety of detection methods including PCR, Lateral Flow, ELISA and plating on selective agar.

Protocols are universal and each cartridge can be run independently, making the system convenient and easy to use. Just load, push the button to start, and walk away.

Supports Cost-saving Pooling Protocols Validated by the AOAC Research Institute
Because the Pathatrix® Auto Instrument efficiently concentrates target bacteria from large volumes, it supports pooling protocols where up to 10 samples may be pooled after enrichment but before downstream detection. Pooling of ten post-enrichment samples reduces downstream detection costs by up to 90%. Pooling protocols validated by the AOAC Research Institute are available for Salmonella, E. coli O157:H7 and Listeria.

Reduces False-Positive and Indeterminate Results
The Pathatrix® Auto Instrument, when used in conjunction with its associated target organism-specific kits, efficiently isolates target organisms away from background flora and potential PCR inhibitors. The result is fewer indeterminate readings due to PCR inhibition and fewer false positives. The system has been successfully used upstream from PCR with 'difficult' matrices such as chocolate, spices, and chicken litter samples, as well as with dozens of other food sample types.

Components
The Pathatrix® Auto Instrument includes 5 Cartridges, a Cartridge Holder, a Sample Vessel Holder and an Elution Vessel Holder. The Elution Vessel Holder includes a magnet to capture paramagnetic particles after elution.

The Pathatrix® Auto is compatible with 100-240V (50-60 Hz) power outlets, and is supplied with three different power cords (NEMA 5-15, BS 1363 and CEE 7/16 Europlug).

Pathatrix™ Cronobacter spp. 10-Pooling Kit Applied Biosystems™

The Pathatrix® Cronobacter spp. 10-Pooling Kit, when used with the Pathatrix® Auto Instrument, facilitates the concentration and isolation of Cronobacter spp. bacteria from pre-enriched food and environmental samples for downstream detection by PCR, immunoassay, or culture plating. This kit format supports pooling of up to 10 enriched samples into a single test, which can help reduce downstream testing costs by up to 90%.

When used with the Pathatrix® Auto Instrument, the Pathatrix® Cronobacter spp. 10-Pooling Kit helps:
• Save up to 90% on downstream detection costs via pooling protocols
• Reduce false positives and indeterminates by separating target bacteria from background flora and inhibitors via a cationic paramagnetic bead

Reduce Cost and Effort

Kit Sensitivity

The limitation of the Pathatrix® Cronobacter spp. 10‐Pooling Kit is in the ability of the target to reproduce in the enrichment medium, be captured by the magnet, and subsequently be detected by either PCR or selective agar.

Pathatrix™ 5 Pooling Listeria spp. Kit Applied Biosystems™

The Pathatrix® 5 Pooling Listeria spp. Kit, when used with the Pathatrix® Auto Instrument, facilitates the concentration and isolation of Listeria spp. bacteria from pre-enriched food and environmental samples for downstream detection of Listeria spp. and Listeria monocytogenes by PCR, immunoassay or culture. This kit format supports pooling of 5 enriched samples into a single test, which can reduce downstream testing costs by up to 80%.

When used with the Pathatrix® Auto Instrument, the Pathatrix® 5 Pooling Listeria spp. Kit:

The Pathatrix® Auto Cartridge Holder is designed for use with the Pathatrix® Auto Instrument, and holds a single Cartridge. The holder provides a steady base that simplifies loading and unloading of the plastic consumable kits into and out of the Cartridge.

Pathatrix™ Sample Vessel Holder Applied Biosystems™

The Pathatrix® Sample Vessel Holder is designed for use with the Pathatrix® Auto Instrument. The apparatus holds the Sample Vessel and Elution Vessel and facilitates easy loading of the vessels with the sample and elution solution, respectively. The Elution Vessel holder features a magnet positioned for the rapid capture of the paramagnetic particles.

Pathatrix™ 10 Pooling E. coli O157:H7 Kit Applied Biosystems™

The Pathatrix® 10 Pooling E. coli O157:H7 Kit, when used with the Pathatrix® Auto Instrument, permits the concentration and isolation of E. coli O157:H7 bacteria from pre-enriched food and environmental samples for downstream detection by PCR, immunoassay or culture. This kit format supports pooling of 10 enriched samples into a single test.

When used with the Pathatrix® Auto Instrument, the Pathatrix® 10 Pooling E. coli O157:H7 Kit:

Pathatrix™ 10 Pooling Kit Applied Biosystems™

The Pathatrix® 10 Pooling Kit simplifies the measurement and pooling of samples from 10 individual pre-enriched food and environmental samples. The single-use sterile straws maintain pre-enriched sample integrity for the subsequent analysis of any positive sample. Each kit includes 100 sterile 10 ml syringes and 1000 sterile 254 mm straws combined into convenient packs of 50. Use this kit with the Pathatrix® Foam Filters (available separately) to remove particulate matter from samples prior to pooling.

Pathatrix™ Cronobacter spp. 5-Pooling Kit Applied Biosystems™

The Pathatrix® Cronobacter spp. 5-Pooling Kit, when used with the Pathatrix® Auto Instrument, facilitates the concentration and isolation of Cronobacter spp. bacteria from pre-enriched food and environmental samples for downstream detection by PCR, immunoassay, or culture plating. This kit format supports pooling of up to 5 enriched samples into a single test, which can help reduce downstream testing costs by up to 80%.

The Pathatrix® Auto Cartridge Rack is designed for use with the Pathatrix® Auto Instrument. The rack holds a set of 5 Cartridges and provides a steady base that simplifies loading and unloading of the plastic consumable kits into and out of the Cartridges.

Pathatrix™ Auto Cartridge Applied Biosystems™

The proprietary patented magnetic separation technology with which the Pathatrix® Auto Instrument performs pathogen enrichment and purification occurs within plastic disposable vessels loaded into a Pathatrix® Auto Cartridge. Each Pathatrix Auto Cartridge can be loaded and run independently of the others, and each Pathatrix Auto Instrument has slots for five cartridges.

Contaminated samples are first added to the consumable sample vessel. Paramagnetic beads, surface-modified for pathogen capture, are then added, and the sample vessel and attached elution vessel are loaded into the Pathatrix Auto Cartridge. The Pathatrix Auto Instrument uses magnets housed in the Pathatrix Auto Cartridge to capture the beads, and then efficiently recirculates the sample over the captured beads. Target organisms are retained by the immobilized beads, and after an automated washing and elution step, the isolated organisms are ready for downstream detection. The entire process is automated and takes only about 15 minutes.

Although the Pathatrix® Auto Instrument includes 5 Pathatrix® Auto Cartridges, this additional cartridge may be purchased, either as a replacement, or to provide the added convenience of being able to load a sample while others are running in the instrument.

A Pathatrix® Auto Cartridge Holder (ordered separately) serves as a stable platform for each cartridge during loading and transport.

Pathatrix™ Elution Vessel Holder Applied Biosystems™

The Pathatrix® Elution Vessel Holder is for use with the Pathatrix® Auto Instrument. The device holds 5 Elution Vessels and each position of the rack features a magnet positioned for the rapid capture of the paramagnetic particles.

Pathatrix™ E. coli O157:H7 Kit, same day Applied Biosystems™

The Pathatrix® E. coli O157:H7 Kit – Same Day, when used with the Pathatrix® Auto Instrument, permits the concentration and isolation of E. coli O157:H7 bacteria from pre-enriched food and environmental samples for downstream detection by PCR, immunoassay or culture. The same day format supports protocols that require pre-enrichment for 8 hours or less prior to re-circulation.
