The HT WT PLUS Consumables for Biomek™ FXP Target Prep Express is designed for automation of the target preparation process for expression analysis on Applied Biosystems whole-transcriptome (WT) arrays. Designed with reagent chemistries identical to Applied Biosystems GeneChip WT PLUS Reagent Kit, this high-throughput (HT) solution allows the user to generate hybridization-ready targets from input RNA with minimal hands-on time. Used in combination with the Beckman Coulter™ Biomek™ FXP Target Prep Express (TPE) System, HT WT PLUS Kit is capable of processing up to 4x24 or 4x96 reactions in a single run.

The GeneTitan™ Hybridization, Wash, and Stain Kit for 3'-IVT Arrays contains pre-formulated solutions to process 3'-IVT array plates on the GeneTitan™ System. The reagents are formulated into the fewest number of individual components possible, reducing preparation and handling steps. With enhanced performance, fewer required reagents, and a simple protocol, target preparation has never been so easy.

The GeneTitan Hybridization, Wash, and Stain Kit for 3'-IVT Arrays includes two boxes:

The first box contains sufficient amounts of the hybridization and stain components to process one 96-array plate, four 24-array plates, or six 16-array plates. The hybridization components include the solutions required for preparation of the hybridization cocktail containing the target. The stain components include Stain Cocktail 1 and 3, Stain Cocktail 2, and the Array Holding Buffer. These solutions are used during the processing of the arrays in the GeneTitan System.

The second box includes Wash Buffer A and Wash Buffer B, also required for the processing of the arrays. There is sufficient Wash Buffer A and B to process one plate in any format. For each additional plate that is processed, an additional Wash Buffer A and B module must be purchased.

The Stain Cocktail 1 and 3, Stain Cocktail 2, Array Holding Buffer, and Wash Buffers A and B are ready-to-use solutions. The hybridization cocktail is prepared by combining the Hybridization Mix and nuclease-free water with the target prepared for the specific target labeling assay (IVT or WT).

The GeneTitan Hybridization, Wash, and Stain Kit for 3'-IVT Arrays should always be stored at 2 to 8° C.

Uncover biology hidden in heterogeneous samples
Traditional genome-wide expression analysis requires RNA extracted from samples composed of a relatively large number of cells. However, the gene expression measurements derived from these samples represent averages that can potentially mask key differences between subpopulations of cells within the samples. To better understand the biological significance of these differences in gene expression, researchers are increasingly focused on characterizing and analyzing subpopulations of cells.

Modern technologies, including fluorescence-activated cell sorting (FACS) and laser-capture microdissection (LCM), enable the characterization and subsequent isolation of small populations of cells. However, due to the limited number of cells, these samples typically produce insufficient RNA to obtain robust expression data from microarrays or RNA Sequencing. The 3' IVT Pico Kit, developed specifically to process limited amounts of input RNA, enables researchers to perform robust gene-level analysis on 3' IVT arrays, including GeneChip Human Genome U133 Plus 2.0 Arrays, using just a few cells from a variety of sample types.

A single kit for multiple sample types
Studies often require the analysis of samples derived from multiple types of tissues. Cancer research, for example, commonly involves the comparison of samples derived from fresh frozen tissues and from FFPE tissues. Usually, it is necessary to use different kits to process different sample types, which increases the complexity of the experimental workflow and introduces unwanted variation into the data. The 3' IVT Pico Kit, which can be used to process the majority of sample types used in basic and translational research, streamlines the experimental workflow, and eliminates variability due to different target preparation methods.

Maximize the value of precious samples
Many samples, such as those derived from biopsies, are highly precious because they are unique or otherwise irreplaceable. When working with these precious samples, researchers must focus on preserving the maximum amount of each sample for future experiments. Working with 3' IVT Pico Kit, researchers can use as few as 10 cells to generate robust and reproducible whole-transcriptome data from microarrays, which ensures that the majority of the sample is preserved for future use.

*The recommended amount of input RNA is 500 pg and the minimum amount is 100 pg. Using less than 500 pg of input RNA may result in a slight loss of assay sensitivity. The GeneChip™ Hybridization Control Kit is required for use with GeneChip eukaryotic expression arrays

Each kit contains 20X Hybridization Control Stock composed of pre-mixed biotin-labeled bioB, bioC, bioD and cre, in staggered concentrations, ready to be added directly to the hybridization cocktail. These controls facilitate monitoring of the hybridization process for troubleshooting.

In addition, this kit also contains Control Oligo B2, used to provide alignment signals for image analysis. (Additional Oligo B2 can be ordered separately.)

All kit components have been thoroughly tested to ensure they meet our high-quality standards.

About Poly-A controls
Poly-A controls (lys, phe, thr, and dap) monitor the target labeling process from start to finish. They serve as sensitivity indicators of target prep and labeling efficiency. This kit contains four exogenous, pre-mixed, poly-adenylated prokaryotic controls that are spiked directly into RNA samples before target labeling. Their resultant signal intensities on Applied Biosystems GeneChip Arrays serve as indicators of the labeling reaction efficiency.

About Hyb controls
The 20X Eukaryotic hybridization controls are comprised of a mixture of biotinylated and fragmented cRNA of bioB, bioC, bioD, and cre in staggered concentrations, ready to be added directly to the hybridization cocktail. These controls facilitate monitoring of the hybridization process for troubleshooting. Control Oligo B2 is included to provide alignment signals for image analysis.

HT WT Terminal Labeling and Controls Kit (Applied Biosystems™)

GeneChip™ Poly-A RNA Control Kit (Applied Biosystems™)

The GeneChip™ Poly-A RNA Control Kit is used with the Prokaryotic Target Labeling Assay and the Prokaryotic Target Labeling Assay for 3' expression studies with prokaryotic GeneChip Brand Arrays. The assays utilize reverse transcriptase and random hexamer primers to produce DNA complementary to the RNA. The cDNA products are then fragmented by DNase I and labeled with terminal transferase and biotinylated GeneChip DNA Labeling Reagent at the 3' termini.

Functionally tested on GeneChip Brand Arrays, we recommend the Prokaryotic Target Labeling Assay and associated reagents for use in prokaryotic gene expression studies.
