As SARS-CoV-2 continues to spread around the globe, COVID-19 testing remains a cornerstone for helping to control the spread of this novel coronavirus. In this workshop, ARUP Laboratories will share their experience using the Emergency Use Authorized TaqPath COVID-19 Combo Kit. They will share clinical performance and comparison data with two other SARS-CoV-2 real-time-PCR based tests, including performance data with regards to the “two of three” infected patient gold standard. Additionally, Thermo Fisher Scientific will discuss our rapidly expanding COVID-19 testing portfolio which aims to assist with the growing testing demands of the medical community.

In this workshop, our customer and collaborator, Dr. Dan Liebler, President of Protypia, Inc., will highlight his research on MS proteotyping of human Cancers in oncology therapeutic development using Thermo Scientific™ Orbitrap™ technology.

New therapeutics targeting immune checkpoint proteins have significantly advanced treatment of non-small cell lung cancer (NSCLC), but protein level quantitation of drug targets presents a critical problem.

Dr. Liebler focuses on his use of multiplexed, targeted mass spectrometry (MS) to quantify immunotherapy target proteins PD-1, PD-L1, PD-L2, IDO1, LAG3, TIM3, ICOSLG, VISTA, GITR, and CD40 in formalin-fixed, paraffin-embedded (FFPE) NSCLC specimens. Global proteome analyses identified distinct proteotypes associated with high PD-L1-expressing and high IDO1-expressing NSCLC.

Part 1: The Thermo Scientific Tox Explorer Collection: Robust LC-MS Workstream for Toxicology Analysis

This presentation will demonstrate a targeted screening and quantitation method for a large panel of drugs of abuse. This workstream based approach utilizes the high-resolution, accurate-mass Thermo Scientific™ Q Exactive™ Plus Orbitrap mass spectrometer. Identified drugs are screened against an extensive HRAM MS/MS spectral library and a database containing molecular formula, exact mass, retention times, and fragment ions.

Part 2: Thermo Scientific TraceFinder™ Software Overview and Implementation of a Thermo Scientific™ Q Exactive™ Hybrid Quadrupole-Orbitrap™ Mass Spectrometer for the Identification and Quantitation of Synthetic Cannabinoids in Postmortem Forensic Investigations

Synthetic cannabinoids continue to appear in forensic toxicology casework, and they represent a chemically diverse class of substances with particularly difficult behaviors to which high resolution mass spectrometry (HRMS) can provide great benefit. A quantitative confirmatory test was developed and validated for 5F-MDMB-PICA (the most prevalent synthetic cannabinoid identified in forensic casework) and its metabolites in human blood. A Thermo Scientific™ Q Exactive™ Hybrid Quadrupole-Orbitrap™ Mass Spectrometer was used for analysis. This interactive presentation will provide an overview of TraceFinder software along with a review of the performance characteristics to demonstrate the utility and accuracy of the developed assay.
